I emailed the developer and he suggested checking the "Open in 32-bit mode" option in the Get Info window for Safari. I cleared various Safari enhancements and reinstalled DownloadComment 1.2.1, did the Get Info thing, and it's working again on my (intel) MBPro, 10.6.4, Safari 5.0.2. I haven't finished upgrading all my Safari apps to discover if there are any conflicts with running in 32-bit mode, but at least I know how to make it work when it's crucial.
